Analysis of bioavailable toluene by using recombinant luminescent bacterial biosensors with different promoters

Abstract In this study, we constructed recombinant luminescent Escherichia coli with T7, T3, and SP6 promoters inserted between tol lux genes as toluene biosensors evaluated their sensitivity, selectivity, specificity for measuring bioavailable in groundwater river water. The luminescence intensity of each biosensor depended on temperature, incubation time, ionic strength, concentrations coexisting organic compounds. Toluene induced the highest -expressing E. T7 promoter [T7- lux-E. , limit detection (LOD) = 0.05 ?M], followed by that T3 (T3- LOD 0.2 ?M) (SP6- 0.5 ?M). Luminescence may have been synergistically or antagonistically affected compounds other than toluene; nevertheless, low benzoate analogs had no such effect. reproducibility experiments, relative standard deviation (4.3–5.8%). SP6- demonstrated high adaptability to environmental interference. T7- biosensor—with LOD, wide measurement range (0.05–500 ?M), acceptable (? 14.3 9.1%)—is an efficient biosensor. This is first study evaluating different potential application actual water bodies.
